We show here that vagal afferent fibres of the caudal ventrolateral medulla (CVLM) project to the paraventricular nucleus of the hypothalamus (PVN) in rats and were excited by flavour-associated responses at the tongue. Furthermore, this excitatory CVLM-PVN projection was abolished by the NMDA-receptor antagonist MK-801 and mimicked by iontophoretic application of NMDA to the CVLM of anaesthetised rats. The excitatory CVLM-PVN projection was selectively sensitive to NMDA while receptors were similarly sensitive to AMPA in cortical areas, including primary gustatory cortex, or in the core of gustatory nucleus. The projection also received a strong cholinergic input and was abolished by atropine. The functional significance of the excitatory CVLM-PVN projection, which is likely to involve NMDA receptors, is unclear. However, we suggest that it may be involved in integrating taste-mediated signals to the brain to regulate energy balance and that blocking this projection may provide a novel means of controlling food intake.[The use of steroids in concomitant infiltrating thyroid carcinoma and metastases].
